Abstract
Abstract in Deutsch:Mit steigenden Titern durch kontinuierliche Produktion und optimierte Prozesse wird die Aufreinigung zum Engpass in der biopharmazeutischen Industrie. Ein kontinuierlicher Aufreinigungsprozess mit hohem Durchsatz wird benötigt. Das Ziel dieser Arbeit war daher die Entwicklung eines neuartigen Aufreinigungsprozesses ohne Chromatographie Säulen. Im Rahmen dieser Arbeit wird das Konzept eines vollkontinuierlichen Reinigungsprozesses vorgestellt, bei dem ausschließlich 3D-gedruckte Hydrozyklone, eine Pumpe und ein Rührer zum Einsatz kommen. Der Hydrozyklon mit 8 mm Durchmesser hat Kapazitäten mit einem volumetrischen Durchsatz von mehr als 0,5 L pro Minute, niedrige Rückdrücke im Vergleich zur Chromatographie und kein Potential für Fouling. Die Konstruktion des Hydrozyklons basiert auf Designs anderer Autoren und wurde entsprechend dem Verwendungszweck angepasst. Zur Veranschaulichung des entwickelten Verfahrens wurde 1 L eines Proteingemischs mit grün fluoreszierendem Protein (GFP) und Lysozym erfolgreich aufgetrennt, wobei 42 % GFP gewonnen wurde und Lysozym um 95 % und das Volumen um mehr als die Hälfte in 8 min (ohne Bindungszeit) verringert wurden. Dies wird durch die Bindung von GFP an ein handelsübliches starkes Anionentauscher Chromatographie Harz erreicht, das aus hydroxyliertem Methacrylpolymer besteht und mit quaternären Aminogruppen funktionalisiert ist. Lysozym wird reduziert, indem die geladenen Beads mit einem mittleren Partikeldurchmesser von 65 µm abgetrennt und mit Puffer gewaschen werden. Um aussagekräftige Massenbilanzen aufzustellen und die Trennfähigkeit des Hydrozyklon-Designs zu bestimmen, werden verschiedene Quantifizierungsmethoden für die Harzpartikel untersucht. Dabei ermittelten wir den Verlust von Chromatographie Partikeln mit einem volumetrischen Ansatz während des Prozesses auf weniger als 8 %. Abstract in Englisch:With increasing titres due to continuous production and optimised processes, purification becomes the bottleneck in the biopharmaceutical industry. A continuous purification process with high throughput is needed. Thus, the aim of this thesis was the development of a novel purification process without chromatography columns. Furthermore, a process using rapid prototyping for different demands in production was investigated. Within this thesis the concept of a fully continuous purification process solely employing 3D-printed hydrocyclones, a pump and stirrer is presented. The hydrocyclone with 8 mm in diameter has capacities of a volumetric throughput more than 0.5 L per minute, low pressures in comparison to chromatography and no potential for fouling. The hydrocyclone design is based on literature and further evaluated to fit the purpose. To showcase the developed process, 1 L of a protein mixture with green fluorescent protein (GFP) and lysozyme was successfully separated yielding 42 % GFP while reducing lysozyme by 95 % and the volume by more than half in 8 min (excluding binding time). This is achieved by binding GFP to a commercially available strong anion exchange resin composed of hydroxylated methacrylic polymer and functionalised with quaternary amine groups. Lysozyme is reduced by separating the charged beads with a mean particle diameter of 65 µm and washing them with buffer. Different quantification methods for resin particles are investigated to set up conclusive mass balances and to determine the separation ability of the hydrocyclone design. We determined the loss of chromatographic beads with a volumetric approach during the process to be less than 8 %. As comparison to the hydrocyclone-based process we then modelled a theoretical chromatography process using experimental data. ...
