Abstract in Deutsch:Untersuchung von Arabidopsis Thioninen unter besonderer Berücksichtigung von THI2.4


Thionine sind pflanzliche antimikrobielle Peptide die als Präproproteine, bestehend aus einem Signalpeptid, dem eigentlichen 5 kDa Thionin und einer sogenannten sauren Domäne, produziert werden. Arabidopsis enthält 4 Thionine und ihre Proproteine wurden als Fusionsproteine mit Thioredoxin in E. coli produziert.
Das Gen Thi2.4 wurde im Detail untersucht. Überexpressionslinien hatten eine deutlich erhöhte Resistenz gegenüber Pseudomonas syringae pv tomato DC3000, Botrytis cinerea und dem Rübenzystennematoden Heterodera schachtii. Die Überexpressionslinien wiesen deutlich erhöhte Calloseablagerungen nach Infektion mit Pst D3000 auf. Die Expression von Thi2.4 wurde mit Promotor::GUS Linien untersucht. Die Expression wurde durch Pst DC3000 induziert und war abhängig von Jasmonat.
Das THI2.4 Peptid wurde wie die Proproteine als Fusionsprotein mit Thioredoxin im Zytoplasma von E. coli produziert. Das gereinigte Thionin wurde in vitro getested und zeigte antimikrobielle Aktivität gegenüber Pst DC3000, Hefe und verschiedenen filamentlösen Pilzen. Die höchste Aktivität wurde gegenüber Fusarium-Arten gefunden.
Um die Funktion der sogenannten sauren Domänen in den Thionin-Proproteinen zu untersuchen wurden transgene Linien für die Arabidopsis Thioningene Thi2.1 und Thi2.3 hergestellt. Die Expressionskonstrukte enthielten sowohl die kompletten Präproproteine als auch solche ohne saure Domäne. Ausserdem wurden Konstrukte kloniert welche nur die saure Domäne mit dem jeweiligen Signalpeptid exprimierten. Ein Vergleich dieser transgenen Linien zeigte einen höheren Transkriptlevel für die Linien die nur das Thionin ohne saure Domäne exprimierten im Vergleich mit den kompletten Konstrukten. Für die Analyse der Expression auf Proteinebene wurden Antikörper gegen die Proproteine produziert.

Thionins are plant AMPs which are produced as preproproteins, consisting of a signal peptide, the thionin itself, and C-terminal acidic domain. After processing they give rise to the mature thionin peptide of 5 kDa. Arabidopsis contains 4 thionin genes. In the present study expression of all 4 Arabidopsis thionin proproteins as fusion proteins in the periplasm or the cytoplasm of E. coli was compared. Fusion proteins were successfully produced inside cytoplasm and tested for antimicrobial activity.
One of the Arabidopsis thionin genes, Thi2.4 was studied in detail. It was overexpressed using the 35S CaMV promoter and homozygous lines were developed. The overexpression of Thi2.4 resulted in enhanced resistance against Pseudomonas syringae pv tomato DC3000, Botrytis cinerea and the beet cyst nematode Heterodera schachtii. Overexpression lines resulted in high callose deposition after Pst DC3000 infection than a T-DNA mutant and wild type plants. The expression of Thi2.4 in different tissues was analyzed with GUS fusions. Thi2.4 was found inducible by infection of Pst DC3000. From the JA and SA impaired mutant studies at transcript level after Pst DC3000 infection it was found that Thi2.4 was regulated by JA.
The THI2.4 peptide was expressed as a thioredoxin fusion protein in the cytoplasm of E. coli, digested with TEV protease, and purified by HPLC. The purified THI2.4 was antimicrobial against filamentous fungi under in vitro conditions and was found to cause delayed spore germination, hyperbranching and swelling tips of fungal hyphae. THI2.4 was also toxic to non-filamentous fungi and Pst DC3000 under in vitro conditions.
To determine the function of the acidic domain of thionins, constructs without acidic domain and only acidic domain for Thi2.1 and Thi2.3 were cloned in pPZP3425 and overexpression lines were developed. The overexpression lines resulted in high transcript levels for these constructs. The maximum increase in transcript level was found for the overexpression constructs of the acidic domains of Thi2.1 and Thi2.3. Fusion proteins proTHI2.1-TRX and proTHI2.3-TRX were used to raise antibodies. These antibodies were purified and found functional after testing them with Western blotting.
